Lipid Sciences, Inc. is developing a technology to reverse atherosclerosis. They have developed a vascular lipid removal system, which sytemically removes lipids from plasma lipoproteins. The technology only removes the lipid component of the particle without adversly affecting lipoproteins, leaving the particle intact and functional. Preclinical studies have domonstrated reverse cholesterol transport in animals. A human safety study is currently being conducted and European trials are planned for 2003.

ZOCOR® (simvastatin, MSD), an HMG-CoA reductase inhibitor, is indicated to reduce elevated total and DL cholesterol and triglycerides, and to increase HDL cholesterol in patients with hyperlipidemia. In patients with coronary heart disease, ZOCOR is indicated to reduce the risk of death, coronary death and non-fatal MI; stroke and TIA; revascularization procedures, and to slow the progression of coronary atherosclerosis. As shown in the 4S study, ZOCOR 20-40mg reduced the risk of total death by 30 percent and the risk of coronary death by 42 percent. ZOCOR has been in clinical use for 15 years and has been prescribed for more than 40 million patients worldwide.

Ezetimibe (eh-ZET-i-mibe) is an investigational new drug and, will be when approved for use, the first drug in a new class of cholesterol lowering medications. In clinical trials, ezetimibe plus a statin provided LDL-c lowering of an additional 25% when added to ongoing statin therapy, HDL-c and reduced triglycerides without compromising patient safety.
The distinct mechanism of action of ezetimibe complements the effects of statins. Ezetimibe works by a novel mechanism in the cells that line the wall of the small intestine to selectively inhibit the intestinal absorption of cholesterol. Unlike bile acid sequestrants, ezetimibe does not physically bind bile acids. Ezetimibe is not a resin, fat-blocker, stanol or sterol. In clinical trials, ezetimibe had a side effect profile comparable to placebo, did not induce cytochrome P-450 (CYP450) drug metabolizing enzymes and has no clinically significant drug interactions. Ezetimibe is administered as one tablet taken once daily at any time, with or without meals.
