ESTROGEN BIOSYNTHESIS IN CENTRAL NERVOUS SYSTEM AND PERIPHERY WERE DIFFERENTLY AFFECTED BY APOE GENOTYPE

Clinical and epidemiological studies have conformed that the effection of estrogen replacement therapy (ERT) on the cognition in AD depends on ApoE genotype. Our clinical investigation demonstrated that natural menopause age of ApoE4 carrier women delayed significantly compared with other genotype carriers. Our data from ApoE transgenic mice showed that hippocampus estrogen level was lower in ApoE4-tg mice while the plasma estrogen level was higher than that of ApoE3-tg mice; The relative activity of aromatase in hippocampus of ApoE4-tg mice was lower too; However, aromatase mRNA transcription level was upregulated in ApoE4-tg mice's hippocampus. In ApoE stable transfected C6 cell line model, we found that ERK signal was involved in regulating the aromatase level just in ApoE3 cells. In conclusion, our data show that ApoE4 have different effects on estrogen biosynthesis in central nerve system and periphery, which may be important with respect to the ERT in AD.
